1.On the basis of GPI-1046(a ligand of FKBP12) in complex with FKBP12, a set of 2,2-dimethyl-N-alkyl dioxo-4-thiazolidine carboxylate derivatives was designed, and computer-aided screened by DOCK4.0 based on the structure of FKBP52. 22 of 500 highest score compounds had been synthesized and their neurotrophic actions had been examined.
本论文以FKBPs的配体GPI-1046为基本分子，基于FKBP12的结构设计了一类2，2-二甲基-N-取代二氧代噻唑烷酸酯类化合物，并使用DOCK4.0针对这类化合物的虚拟库基于FKBP52的结构进行计算机辅助药物筛选，合成了评分较高的500个分子中的22个，并进行了体外活性筛选。
